Single Player Game:

Obviously, if playing a single player game, Playing the role of Earth,
Centauri, or Narn will be the biggest challenge.
Minbari will be slightly easier, and Vorlon or Shadows will be by far
the easiest (4 and 5 racial advantages).
Make further adjustments to suit your playing tastes and level of
experience - Take off Durable Ships, fliers and Devious Schemers
(in that order) from Shadows and Vorlons if necessary.
If NOT playing Shadows and you want an even bigger challenge - add
fliers, Expert Builders, etc. to Racial Advantages of Shadows.

Multi Player Games:

2 player - I would recommend playing Vorlons vs Shadows
           (Add Durable Ships to Vorlons)
3 player - Earth, Minbari, Narn.
4 player - Earth, Minbari, Narn, Centauri
5 player - Earth, Minbari, Narn, Centauri, Vorlon
(Computer plays races not listed)

The Shadows are considered the main enemy.
For a co-op style multi-player game against Shadows it may be necessary
to increase the challenge by adding extra Racial Advantages to Shadows.
I would recommend adding fliers, Expert Builders, and elements to increase
expansion early in the game such as Ship Craftsman, Hardy Spacefarrers.
